Antioxidant activity of the extracts from Pycnoporus sanguineus mycelium BABT

The aim of this work was to study the production of compounds with antioxidant activity by Pycnoporus sanguineus when cultivated in submerged fermentation using a potato dextrose broth plus peptone medium. The study evaluated the biomass production, glucose consumption, variation of medium pH and antioxidant activity. The antioxidant potential was tested through the DPPH method and β-carotene / linoleic acid system with extracts obtained from the mycelium at different times of cultivation (5, 10, 15, 20, 25 and 30 days). Faster chitosan production by mucoralean strains in submerged culture BJM

Production of chitosan was conducted using two Mucoralean strains, Mucor racemosus and Cunninghamella elegans. Chitosan was extracted from mycelia of M. racemosus and C. elegans at different growth phases on YPD medium. In both fungi, chitosan was rapidly produced, while highest yield of extractable chitosan was found in 24h of cultivation in submerged culture. The yield of chitosan isolated from dry mycelia of M. racemosus was about 40% higher than from C. elegans. The degree of N-acetylation of chitosan was 49% in M. racemosus and 20% in C. elegans, and the D-glucosamine contents were about 48% and 90%, respectively.

Optimization of exopolysaccharides production from a novel strain of Ganoderma lucidum CAU5501 in submerged culture BJM

This study aimed at optimizing the medium of a new Ganoderma lucidum strain CAU5501 to enhance the yield of exopolysaccharides (EPS) and mycelial growth. Firstly, the suitable level of glucose, magnesium, phosphate and C/N ratio was determined by single factor experiment. Subsequently, the optimum concentrations of these medium components were investigated using the orthogonal matrix method. The results indicated that the higher levels of EPS were correlated with the level of cell growth when glucose concentration was studied (data no show). The optimum medium for EPS yield was found to be 70 g/l glucose, 5 C/N ratio, 2.5 g/l KH2PO4, 0.75 g/l MgSO4·7H2O, and for mycelial growth was 50 g/l glucose, 5 C/N ratio, 1.5 g/l KH2PO4, 0.5 g/l MgSO4·7H2O. Abstract This work aimed to evaluate the production of lipolytic complexes, produced by microorganisms isolated from a biological treatment system of effluents from a hotel. To select the best lipolytic microorganism for use in biotechnological processes, we tested 45 bacterial isolates recovered from the raw effluent of the hotel’s restaurant waste tank. Lipase production was assayed in culture medium supplemented with olive oil and rhodamine B, incubated at 25 °C and 30 °C for 24 h - 48 h. Results showed 22 isolates lipase producers. All isolates were inoculated on medium without yeast extract to select the ones with highest enzyme yields. Out of these, nine isolates showed high lipase activity. Screening of basidiomycetes for the production of exopolysaccharide and biomass in submerged culture Rev. Microbiol.

Fifty-six strains of Basidiomycetes, including native Brazilian fungi isolated from different ecosystems and edible mushrooms, were screened for production of exopolysaccharides and biomass in submerged culture. Agaricus sp. (CCB 280) and Oudemansiella canarii (Jungh.) Hohn (CCB 179) were the highest exopolysaccharide producers (6.01 and 3.54 g dry w./l respectively) after 7 days of incubation. The best producer of biomass was Schizophyllum commune Fr.:Fr. (CCB 473) with 16.68 g dry w./l in 14 days of incubation. When the culture filtrate was submitted to freezing prior to polysaccharide precipitation, a gelatinous fraction was formed.

The selection of new microorganisms able to produce antimicrobial compounds is hoped for to reduce their production costs and the side effects caused by synthetic drugs. Clavulanic acid is a β-lactam antibiotic produced by submerged culture, which is widely used in medicine as a powerful inhibitor of β-lactamases, enzymes produced by bacteria resistant to antibiotics such penicillin and cephalosporin.
